Theory
Binary fission in Amoeba is asexual reproduction where the organism divides into two equal daughter cells after DNA replication. Budding in yeast is asexual reproduction where a small outgrowth (bud) forms on the mother cell, develops, and separates to form a new individual.
Procedure
- Prepare a wet mount of Amoeba culture by placing a drop on a slide with a cover slip
- Observe under the microscope at 40X magnification
- Look for Amoeba showing stages of binary fission: early stage with dividing nucleus, later stage with furrow formation, and two separate daughter cells
- For yeast, prepare a wet mount of yeast culture
- Observe yeast cells showing budding: small bud emerging from mother cell, bud growing, and bud separating
- Record observations of different stages of reproduction
- Draw labeled diagrams of the observed stages
Observation
Amoeba cells in different stages of binary fission are observed. The nucleus divides, and the cytoplasm cleaves to form two identical daughter cells. Yeast cells show different stages of budding with small buds emerging from mother cells and larger buds ready to separate.
Result
Both asexual reproduction processes are successfully observed. Amoeba reproduces by binary fission producing two equal cells, while yeast reproduces by budding producing unequal cells (small bud and large mother cell).
